Usually when earpiece audio is out, it is a faulty Flex Cable #3. Less often it is the speaker itself. So most start with trying to reseat the Cable #3 to see if they can get it working, if not they then replace Cable #3. At the time of replacement you can even check to see if there is voltage across the speaker with a voltage meter showing it is still probably OK. If still not working there may be damage to Connector #3 on the Logic Board, or the speaker is really out. During repairs of 3GS make sure iPhone is OFF.

Yes proximity sensor works.

I replaced the entire glass,LCD assembly including a (working ) speaker. I seems it must be the connector or something else on the logic board.

There are plenty of vendors out there that sell complete screen assemblies with #^$* poor quality parts. I would bet that it is your ribbon that is causing the ear piece to not work. It can still allow the proximity sensor to work without a functioning ear speaker. Ive seen this way too many times. Replace the ribbon with a quality one and report back.

I recently had this issue after replacing the wifi antenna. I broke the clip that locks the ear speaker flex cable in place. I was able to fix it by placing 3 tiny sheets of paper on the bottom of the flex cable and then pushing into the slot. Not sure how long it will last, but it works!
