I deleted my hard drive icon?

I was attempting to back up my iMac to an external hard drive so that I could instal the additional RAM required to run Snow Leopard. Somehow, the hard drive icon went "poof" and disappeared. Now when I turn it on, I just get a flashing ? folder. When I try to reinstall the software, when I get to select destination, it gives me no choices? Help?

If, when starting up from your system installation disk and in Disk Utilities you don't see the hard drive icon on the left, your hard drive has died. Please give us the last three letters of your serial number so we can identify your machine and direct you to the correct repair guide.

Oh no...will I have to replace my hard drive? Will I lose everything or can I recover it somehow???

If Disk Utilities will not see it, It's probably gone. What were you using to try to back it up?
