I just did an experiment with my 3GS phone and if I power it OFF and plug it into the PC via the standard iPod cable it powers up. I can then power off with switch as normal

Sounds like the headphone assy (includes power switch) or button are faulty.

That would fit your description - cant turn on, cant turn off when connected to iPod cable.

So have you replaced the switch (headphone assy)? What happens if you press the home button while in the current state?
