This can be caused by a number of issues, ranging from software to hardware. Try resetting the phone first by holding both buttons for 10 seconds. If it's still not responding you have a hardware issue.

1) the contacts under the home button are not touching their receiving points

2) the home button flex cable button has failed and needs to be replaced.

3) surface contacts on the dock/speaker/mic/antenna assembly may be worn or corroded

4) In RARE circumstances this can be mainboard related (usually only occurs after a serious drop)

You can use the teardown manuals here on iFixIt to open the phone, check and clean the contacts, and test again. Replacing the home button requires removing and (if you care about your phone) replacing the touchscreen glass. Replacing the dock connector is easier, but generally is not the issue.

try turning off your iPhone then turning it back on, or popping the battery out & then putting it back. Works with my Nokia E71. and it beats trying to reinstall the phone's software. YMMV

Please let me know if that worked. In the Nokia E71 it seems to be a memory problem.
