Optical Bay for iMac 27¨ mid 2010
Does this Optical Bay, iMac & Mac mini Dual Drive Enclosure, also work with an iMac Intel 27-inch mid 2010 (EMC 2390)?

You don't need to give up your optical drive in this system. This system has a spare SATA port to support a second drive. While Apple didn't offer it in this series it was able to support an additional drive (SSD).

But I'm actually only interesting if this Optical drive will work on my iMac as well, since I don't have any need for the optical drive.
Are you looking at three drives then ;-} Sure you can use it to hold a third drive.
