Plug it into your computer and start iTunes. When you select the iPod from the list of libraries on the left, there should be a button to restore to factory settings. That will wipe everything from the iPod.

As this was posted in 2010, a smol update is at hand: Select the iPod icon under the menu buttons on the top left corner > Look for the "Restore iPod" button, click it. It will prompt you with a message asking if you'd like a Factory Reset (which erases all media as well)

I've had that happen a few times on Windows. Try this:

1) Hold menu and select on your iPod until the Apple logo appears.

2) Exit iTunes.

3) In task manager, kill iPod service.

4) Open iTunes.

5) Connect iPod.

Seems to help mine when iTunes won't detect it.
