Pressure spots after replacing screen

So I just (sort of) completed my first screen replacement for an iPod Touch 4G. Taking it apart and putting it back together was pretty easy, but now that it's back together, the LCD is giving pressure spots whenever I press somewhere even moderately hard (I'm not being very forceful, no more force than most people use when holding and typing on the iPod). I read somewhere that the screws might not be screwed in all the way but I already tightened them twice. Any suggestions?

LCD's in general can't handle any pressure contact without creating wells.

Not much you can do here. You could try using a hair dryer to heat up the screen a little bit (over doing it will hurt it as well) that may thin the liquid a bit to flow back if you don't have a pinch point. Otherwise it's a new screen.
