Does the slide to power off screen appear when you hold the power button while the iPad is on or unlocked?

If yes then the issue is linked to the home button flex. I don't remember how the home button is set up for iPad Mini 4 but it it should have touch ID and hall effect sensor chip (that controls smart cover functionality, if it's broken the iPad thinks the cover is closed and disables the lock button press) on there which may need to be transferred from the old home button to the new one or if it's separate you should be able to transfer from old digitizer to new digitizer.

Had the same problem before i noticed the small cable that needs to be soldered onto the new screen. It has 6 connection points, and after transferring and soldering the cable, the on/off button worked perfectly.

Sounds like there is a possibility the cable for the sleep/wake button is not connected properly. Could you please send me the model number? Thanks!

When you put the device back together, (assuming you changed the screen) did you double, triple check the connectors? Made sure none of them were damaged in the opening process? If there is no visble damage, I would suggest a power button replacement.

The new screen must have sleep wake sensor, otherwise power button will not work. Check https://youtu.be/r2pwiSmUB30
