Which RAM slot do I use when upgrading?

My MacBook Core 2 Duo currently only has 1 G of RAM. I'm upgrading to 2 G of RAM. The 1 G had a card in each slot, but the 2 G comes as only 1 card. Which slot do I use? Do I need to put something in the other slot? Is there anything else I need to be aware of when completing this upgrade?

Options

Use either slot, maybe both... some of these machines could run up to 3GB RAM.
