Will the iPad turn on while it is opened?

I want to replace the dock connector and test if it is working before fully reassembling the device again.

Will the iPad (1st gen.) turn on while it is opened (but all cables connected) or is it necessary to fully reassemble the device in order to turn it on again?

In order to have all the cables connected, the only thing not reassembled in your iPad would be the display clips. I don't see why it wouldn't work, as long as you make sure that all cables are connected. I've been able to do that on a Nintendo Wii, but had an iMac beep at me when the EyeSight camera was unplugged.
