I haven't found this really necessary in a long time. I do try to defragment and optimize once a year.

No, you can't defragment a system from an app. that's running on it. You boot from the TTP disk (it has a system file) and then defragment the drive.
1. Remove Unnecessary files from the desktop
Every time the Mac OS X loads, it indexs desktop contents. creates icons for files and catalogs the contents of each folder at desktop.
2. Say “NO” to unwanted startup applications
Reduce the number of startup applications go “System Preferences” -> “User Accounts”. Uncheck those Mac startup applications from the list which you don't need to run at Mac startup. The unnecessary startup applications increase the boot time and slow down your Macbook.
3. Disable the Dashboard – An important fix for slow Macbook
If you are regular user of Mac OS X dashboard then it should be kept in mind that each one of those unnecessary widgets and web clips consumes memory - and you can't even see them unless you ask for them!
5. Disable/Switch Off the unused options in your Macbook
The options like Wireless, Bluetooth, Speech Recognition, Internet Sharing etc. should be turned off when they are not in use. This will not only speedup your Macbook but, extend your battery life too.

open terminal > type:
defaults write com.apple.dashboard mcx-disabled -boolean YES
return
killall Dock
return - exit terminal
You have to log out or reboot to have it take effect.
To turn back on type NO in place of YES
