Galaxy s6 wont power up . Everything internal is connected but I have not connected the screen with adhesive. Does the screen have to be attached for the phone to power up?

No, it doesn’t have to be connected. However, it is difficult to power on when it’s not in the mid frame because of the way the power button was designed. Make sure the board is making full contact with the power button connectors in the mid frame and it doesn’t hurt to connect the screen so you can see if you’re getting a response when you press the power button.
