My MacBook Pro 15" Unibody Mid 2009 with 3.06 GHz Intel Core 2 Duo has 4 GB of memory. If I want to add another 4 GB of memory, do I need to buy two 4 GB cards so that they are compatible or can I just buy one additional 4 GB card?

Yes, you can add just one 4 GB stick at a time. This should give you 6 GB.
You do lose a little bit of speed due to lack of interleaving but it's OK.

Since you MacBook Pro has two RAM slots you will need to get two RAM sticks. So you will need to buy two 4GB cards to get a total of 8GB or RAM.
