I recently replaced the earpiece speaker in my iPhone 4S and everything went well but now the mic doesn't work when I talk on speakerphone only. I am guessing I will need to replace either the power and sensor flex cable or the headphone jack cable, but I don't know which one contains the external mic?

You need to replace headphone jack cable in this case.
You can confirm that microphone used for talk on loudspeaker is broken by recording a video. See if it records any sounds. If nothing happens or it makes noise - replace headphone jack cable
