Can we install 2 disk drives in iMac 2012?
As we can see,there is a SSD port on iMac 2012's logic board.But can we install a SSD and a hard disk drive(not using Fusion Drive) at the same time?

It is a SDXC SD Card slot - not a SSD HD drive slot, that enables these computers to read and write data to SD media inserted in the slot - not use it as a boot drive. You could, store files on it.
